Filho de Diogo Gonçalves Salema I e Leonor Anes. Casado com Catarina Salema. Não teve filhos. Morador na Quinta de Arapouco em Alcácer do Sal. Fidalgo da Casa d’el Rei e da Casa do Infante D. Luís. Casado com Catarina Salema. A primeira referência Rui Salema data de 1537 e encontra-se no contrato do seu casamento. A última referência, datada de 1578, surge na carta patente que confirmou Rui Salema e a sua mulher como fundadores do Mosteiro de Nossa Senhora de Aracoeli em Alcácer do Sal.
