Filha de António da Mota de Almada. Casada com Fernão Gomes da Gama. Teve pelo menos 12 filhos, a saber, Fernão Gomes da Gama, que sucedeu, e outros 2 homens e 9 mulheres. . A primeira referência a Maria de Almada data de 1584 e surge numa cópia não autenticada e sem data do testamento da sua tia paterna, Antónia de Almada, que nomeia a sua sobrinha como herdeira. A última referência, datada de 1590, encontra-se no inventário que fez com o seu marido dos bens com os quais iam dotar uma das suas filhas.
